Leukotriene C4 and D4 formation by particulate enzymes   总被引:6,自引:0,他引:6  
The homogenate of rat basophilic leukemia cells, when incubated with arachidonic acid, glutathione, and calcium, formed 3 isomers of 5,12-dihydroxyeicosatetraenoic acid and 2 isomers of 5,6-dihydroxyeicosatetraenoic acid, as well as leukotriene (LT) C4 and D4. The products were identified by high pressure liquid chromatography, ultraviolet spectral analysis, co-migration with standards, bioassay, and gas chromatography-mass spectrometry. The enzymes responsible for the formation of LTC4 and LTD4 from LTA4 were found in the 10,000 x g pellet and, therefore, appear to be particulate. The possibility that these enzymes are bound to the cell membrane suggest that the formation of these leukotrienes might be important in the basophil and mast cells release reaction.  相似文献

Prostaglandins A2, B1, E1, E2, F1alpha and F2alpha were added to cultures of human epidermal cells (keratinocytes) for 24 hours at 37 degrees C, and the effects on 3H-thymidine uptake into DNA was measured. At 70 mu/ml all prostaglandins tested except PGF2alpha inhibited the uptake of 3-thymidine greater than 50%. However, at 35 mug/ml, PGA2 and PGB1 were the only two prostaglandins to show significant inhibition, 96% and 51% respectively. At 17.5 mug/ml only PGA2 caused substantial inhibition, 68%. In order to determine if the PGA2 action was mediated by membrane receptors propranolol, phentolamine, metiamide and prostynoic acid were added in conjunction with PGA2. None of the above receptor antagonists were able to reduce the PGA2-induced inhibition of 3H-thymidine uptake. These results indicate that the pre-incubation of human keratinocytes with prostaglandins for 24 hours results in a decrease of 3H-thymidine incorporation in DNA. The precise mechanism of action is unknown at this time.  相似文献

Congenital central hypoventilation syndrome (CCHS) patients show impaired ventilatory responses to CO2 and hypoxia and reduced drive to breathe during sleep but retain appropriate breathing patterns in response to volition or increased exercise. Breath-by-breath influences on heart rate are also deficient. Using functional magnetic resonance imaging techniques, we examined responses over the brain to voluntary forced expiratory loading, a task that CCHS patients can perform but that results in impaired rapid heart rate variation patterns normally associated with the loading challenge. Increased signals emerged in control (n = 14) over CCHS (n = 13; ventilator dependent during sleep but not waking) subjects in the cingulate and right parietal cortex, cerebellar cortex and fastigial nucleus, and basal ganglia, whereas anterior cerebellar cortical sites and deep nuclei, dorsal midbrain, and dorsal pons showed increased signals in the patient group. The dorsal and ventral medulla showed delayed responses in CCHS patients. Primary motor and sensory areas bordering the central sulcus showed comparable responses in both groups. The delayed responses in medullary sensory and output regions and the aberrant reactions in cerebellar and pontine sensorimotor coordination areas suggest that rapid cardiorespiratory integration deficits in CCHS may stem from defects in these sites. Additional autonomic and perceptual motor deficits may derive from cingulate and parietal cortex aberrations.  相似文献

Among the materials excavated by the 1975 joint USSR–USA team in Siberia are two burials from Shaman's Cape, Olkhon Island, Lake Baikal. One is a middle-aged male of the Serovo culture, 6,000 B.C. , and the other is a young male of the Glaskovo culture of 2,000 B.C. This later burial displays an unusual pathology affecting the nose and post-cranial regions of the pelvic girdle and lower limbs. Osteon analysis confirms the determination of age at death and illustrates the difference between normal and pathological bone. Numerous cultural materials were associated with these burials, including harpoon heads, knives, a compound fishhook and a pestle with the Serovo man, and nephrite ornaments with the Glaskovo man. The skulls, though far apart in time, are pronouncedly Mongoloid and alike in their low cranial vaults. A low, broad and inclined ascending ramus resembles Chukchi, Eskimos and Aleuts. These two specimens document the Mongoloid character of the early inhabitants of Lake Baikal.  相似文献
